Doudna pel desenvolupament de la t\u00e8cnica CRISPR-CAS9, un m\u00e8tode per a l&#8217;edici\u00f3 de l&#8217;genoma.<\/span><\/strong><\/p>\n<p><!--more--><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">CRISPR-CAS9, descoberta en 1993 per <strong>Francisco Mart\u00ednez Mojica<\/strong>, \u00e9s una t\u00e8cnica de &#8216;talla-enganxa&#8217; gen\u00e8tic, que permet eliminar fragments d&#8217;ADN defectuosos o indesitjats i substituir-los per altres d&#8217;inter\u00e8s.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">&#8220;Aquest any el Premi va sobre reescriure el codi de la vida&#8221;, anunciava el secretari general de la Reial Acad\u00e8mia de les Ci\u00e8ncies de Su\u00e8cia, <strong>G\u00f6ran K. Com va dir Hanson just abans de comunicar el nom de les guanyadores: &#8220;gr\u00e0cies a la seva troballa s&#8217;ha aconseguit reescriure el codi de la vida&#8221;.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">\u00c9s sens dubte un Premi merescut, encara que per a molts cient\u00edfics, falti un nom entre les designades.\u00a0<\/span><\/p>\n<p style=\"text-align: left;\">\u00a0<\/p>\n<p style=\"text-align: left;\"><strong><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Emmanuelle Charpentier<\/span><\/strong><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Emmanuelle Charpentier (Juvisy-sur-Orge, Fran\u00e7a; 1968) va estudiar Bioqu\u00edmica i Microbiologia a la Universitat Pierre i Marie Curie de Par\u00eds i, el 1995, es va doctorar en Microbiologia a l&#8217;Institut Pasteur.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Des de 1996 fins 2002 amplia la seva formaci\u00f3 a la Universitat Rockefeller de Nova York, al Langone Medial Center de la Universitat de Nova York, a l&#8217;Institut Skirball de Medicina Biomolecular de la mateixa ciutat i al St. Jude Children &#8216;s Research Hospital de Memphis.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Posteriorment, va establir el seu propi grup de recerca en els Laboratoris Max F. Perutz de la Universitat de Viena i m\u00e9s tard va obtenir la pla\u00e7a de directora de recerca de l&#8217;Laboratory for Molecular Infection Medicine Sweden (MIMS) de la Universitat d&#8217;Ume\u00e5 (Su\u00e8cia), on era, a m\u00e9s, professora visitant al Centre Ume\u00e5 per a Investigaci\u00f3 Microbiana.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">El 2013, s&#8217;incorpora com a professora en el Helmholtz Centre for Infection Research de Braunschweig (Alemanya) i, el 2014 obt\u00e9 una c\u00e0tedra &#8216;Alexander von Humboldt&#8217; a l&#8217;Escola de Medicina de Hannover (Alemanya).<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Des de 2015 dirigeix el nou Institut Max Planck de Biologia de la Infecci\u00f3 a Berl\u00edn, mantenint la seva pla\u00e7a de professora visitant a la Universitat d&#8217;Ume\u00e5.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Fuster \u00e9s m\u00e9s coneguda pel seu paper en desxifrar i aplicar, com a eina d&#8217;enginyeria gen\u00e8tica, els mecanismes moleculars de sistema immunol\u00f2gic bacteri\u00e0 CRISPR-CAS9 (clustered Regularly Interspaced Short Palindromic Repeats), terme encunyat pel microbi\u00f2leg Francisco Mojica, qui va descobrir aquest fenomen biol\u00f2gic el 1993.<\/span><\/p>\n<p style=\"text-align: left;\">\u00a0<\/p>\n<p style=\"text-align: left;\">\u00a0<\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\"><\/span><\/p>\n<p style=\"text-align: left;\">\u00a0<\/p>\n<p style=\"text-align: left;\">\u00a0<\/p>\n<p style=\"text-align: left;\"><strong><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Jennifer A. Doudna<\/span><\/strong><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Jennifer A. Doudna (19649), va obtenir la seva llicenciatura en Qu\u00edmica a la Universitat de Pomona (Claremont, Calif\u00f2rnia) el 1985, i el seu doctorat en Bioqu\u00edmica sobre ribozims, a la Universitat de Harvard, sota la supervisi\u00f3 de Jack W. Szostak.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Entre 1989 i 1994, va realitzar les seves investigacions postdoctorals a l&#8217;Hospital General de Massachusetts, la Harvard Medical School i la Universitat de Colorado Boulder amb Thomas Cech.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Entre els anys 1994 i 2002, va ocupar diferents posicions en el Departament de Biof\u00edsica i Bioqu\u00edmica Molecular de la Universitat de Yale, l&#8217;\u00faltima d&#8217;elles com a titular de la &#8216;C\u00e0tedra Henry Ford II&#8217; (2000-2002).<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Per estar m\u00e9s a prop de la seva fam\u00edlia i de l&#8217;sincrotr\u00f3 en el Laboratori Nacional Lawrence Berkeley, el 2002, va acceptar la posici\u00f3 com a membre de la facultat de la Universitat de Calif\u00f2rnia, Berkeley, on en l&#8217;actualitat \u00e9s catedr\u00e0tica dels departaments de Qu\u00edmica i de Biologia molecular i Cel\u00b7lular.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Tamb\u00e9, a la mateixa Universitat, \u00e9s responsable de el Centre Li Ka Shing dedicat a Ci\u00e8ncies Biom\u00e8diques i de la Salut i directora executiva de la Iniciativa Innovative Genomics.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Ha estat investigadora a l&#8217;Institut M\u00e8dic Howard Hughes (HHMI) des de 1997, i des de 2018 \u00e9s la investigadora principal en els Instituts Gladstone.<\/span><\/p>\n<p style=\"text-align: left;\">\u00a0<\/p>\n<p style=\"text-align: left;\"><strong><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">La seva recerca<\/span><\/strong><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">El treball inicial que va realitzar en la soluci\u00f3 de grans estructures d&#8217;ARN la van portar a fer m\u00e9s estudis estructurals de la ribozima de virus d&#8217;hepatitis delta (HDV), l&#8217;IRES i complexos prote\u00efna-RNA com la part\u00edcula de reconeixement de senyal.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">El seu laboratori ara s&#8217;enfoca en una entesa mecan\u00edstica dels processos biol\u00f2gics que involucren l&#8217;ARN. Aquest treball est\u00e0 dividit en tres grans \u00e0rees: el sistema CRISPR, ARN d&#8217;interfer\u00e8ncia i controls traduccionals via micro ARN.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">El 2012 Jennifer A. Doudna i els seus col\u00b7legues van realitzar un nou descobriment que redu\u00efa el temps de treball necessari per editar l&#8217;ADN gen\u00f2mic basat en una prote\u00efna anomenada CAS9, que es troba en el sistema immune del bacteri Streptococcus &#8216;CRISPR&#8217;, i que treballa com unes tisores.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">El descobriment de les tisores gen\u00e8tiques prov\u00e9 de l&#8217;estudi dels sistemes immunes de bacteris i altres microorganismes, el pioner ha estat el cient\u00edfic espanyol Francisco Mojica.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Aquest va observar que els bacteris tenen un sistema de defensa que els permet tallar a trossos qualsevol ADN estrany (per exemple procedent d&#8217;un virus que hagu\u00e9s infectat la c\u00e8l\u00b7lula).<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Va ser el 2012 quan Jennifer Doudna i Emmanuelle Charpentier van demostrar que podien utilitzar el mateix principi per fer canvis espec\u00edfics en l&#8217;ADN d&#8217;una c\u00e8l\u00b7lula.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Quan l&#8217;any 2017 les dues premiades van compartir el Premi Fundaci\u00f3 BBVA Fronteres del Coneixement ** amb el propi Mojica, Doudna va declarar que va comen\u00e7ar a treballar en l&#8217;\u00e0rea de CRISPR despr\u00e9s de llegir un article del cient\u00edfic espanyol.<\/span><\/p>\n<p style=\"text-align: left;\">\u00a0<\/p>\n<p style=\"text-align: left;\"><strong><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Els laboratoris de Jennifer Doudna i d&#8217;Emmanuelle Charpentier<\/span><\/strong><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">En col\u00b7laboraci\u00f3 amb el laboratori de Jennifer Doudna, el laboratori d&#8217;Emmanuelle Charpentier va descobrir com CAS9 podria ser usat per fer talls en qualsevol seq\u00fc\u00e8ncia desitjada d&#8217;un genoma i inserir, suprimir o modificar ADN.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">El m\u00e8tode que van desenvolupar implicava la combinaci\u00f3 de CAS9 amb mol\u00e8cules sint\u00e8tiques de &#8216;ARN guia&#8217; de f\u00e0cil creaci\u00f3. Investigadors de tot el m\u00f3n utilitzen aquest m\u00e8tode per manipular de forma efica\u00e7 l&#8217;ADN de plantes, animals i l\u00ednies cel\u00b7lulars de laboratori.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">Des dels primers mesos de 2015 tant Charpentier com Doudna mantenen un litigi de patents amb el bioqu\u00edmic Feng Zhang, encara que realment es tracta d&#8217;una pol\u00e8mica entre les institucions on treballen els litigants:<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-family: 'trebuchet ms', geneva; font-size: 10pt;\">L&#8217;Institut BROAD de MIT, on treballa F. Zhang i un consorci liderat per la Universitat de Calif\u00f2rnia a Berkeley, on treballa Doudna, al costat de la Universitat de Viena, en la qual est\u00e0 Fuster. Les institucions estan molt pendents de l&#8217;prestigi i diners que hi ha en joc.
